CAB3

phosphopantothenoylcysteine decarboxylase complex subunit CAB3, YKL088W
Subunit of PPCDC and CoA-SPC complexes involved in CoA biosynthesis; subunits of the phosphopantothenoylcysteine decarboxylase (PPCDC) complex are: Cab3p, Sis2p, Vhs3p, while the subunits of the CoA-synthesizing protein complex (CoA-SPC) are: Cab2p, Cab3p, Cab4p, and Cab5p as well as Sis2p and Vhs3p; null mutant lethality is complemented by E. coli coaBC

CAB4

putative pantetheine-phosphate adenylyltransferase, YGR277C
Subunit of the CoA-Synthesizing Protein Complex (CoA-SPC); subunits of this complex are: Cab2p, Cab3p, Cab4p, Cab5p, Sis2p and Vhs3p; probable pantetheine-phosphate adenylyltransferase (PPAT); PPAT catalyzes the fourth step in the biosynthesis of coenzyme A from pantothenate; null mutant lethality is complemented by E. coli coaD (encoding PPAT); widely conserved

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Molecular characterization of the heteromeric coenzyme A synthesizing protein complex (CoA-SPC) in the yeast Saccharomyces cerevisiae.

Olzhausen J, Moritz T, Neetz T, Schueller HJ

Coenzyme A (CoA) as an essential cofactor for acyl and acetyl transfer reactions is synthesized in five enzymatic steps from pantothenate, cysteine and ATP. In the yeast S. cerevisiae, products of five essential genes CAB1-CAB5 (coenzyme A biosynthesis) are required to catalyze CoA biosynthesis. In addition, non-essential genes SIS2 and VHS3 similar to CAB3 are also involved. Using epitope-tagged variants ... [more]

An interaction is inferred between proteins in vitro. This can include proteins in recombinant form or proteins isolated directly from cells with recombinant or purified bait. For example, GST pull-down assays where a GST-tagged protein is first isolated and then used to fish interactors from cell lysates are considered reconstituted complexes (e.g. PUBMED: 14657240, Fig. 4A or PUBMED: 14761940, Fig. 5). This can also include gel-shifts, surface plasmon resonance, isothermal titration calorimetry (ITC) and bio-layer interferometry (BLI) experiments. The bait-hit directionality may not be clear for 2 interacting proteins. In these cases the directionality is up to the discretion of the curator.
