Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Ubiquitin E3 ligase Itch negatively regulates osteoclast formation by promoting de-ubiquitination of TNF receptor-associated factor 6 (TRAF6).

Itch is an ubiquitin E3 ligase that regulates protein stability. Itch-/- mice develop an autoimmune disease phenotype characterized by Itchy skin and multi-organ inflammation. However, the role of Itch in the regulation of osteoclast function has not been examined. We report that Itch-/- bone marrow and spleen cells formed more osteoclasts than cells from WT littermates in response to RANKL, ... [more]
