PDR5

LEM1, STS1, YDR1, ATP-binding cassette multidrug transporter PDR5, L000001365, L000002136, L000002504, YOR153W
Plasma membrane ATP-binding cassette (ABC) transporter; multidrug transporter actively regulated by Pdr1p; also involved in steroid transport, cation resistance, and cellular detoxification during exponential growth; PDR5 has a paralog, PDR15, that arose from the whole genome duplication

KTI10, H(+)-exporting P2-type ATPase PMA1, L000001449, YGL008C
Plasma membrane P2-type H+-ATPase; pumps protons out of cell; major regulator of cytoplasmic pH and plasma membrane potential; long-lived protein asymmetrically distributed at plasma membrane between mother cells and buds; accumulates at high levels in mother cells during aging, buds emerge with very low levels of Pma1p, newborn cells have low levels of Pma1p; Hsp30p plays a role in Pma1p regulation; interactions with Std1p appear to propagate [GAR+]

PCA

A Protein-Fragment Complementation Assay (PCA) is a protein-protein interaction assay in which a bait protein is expressed as fusion to one of the either N- or C- terminal peptide fragments of a reporter protein and prey protein is expressed as fusion to the complementary N- or C- terminal fragment of the same reporter protein. Interaction of bait and prey proteins bring together complementary fragments, which can then fold into an active reporter, e.g. the split-ubiquitin assay.

ATP-binding cassette (ABC) transporters are a ubiquitous class of integral membrane proteins of immense clinical interest because of their strong association with human disease and pharmacology. To improve our understanding of these proteins, we used membrane yeast two-hybrid technology to map the protein interactome of all of the nonmitochondrial ABC transporters in the model organism Saccharomyces cerevisiae and combined this ... An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner is identified by mass spectrometric methods.

Co-fractionation

Interaction inferred from the presence of two or more protein subunits in a partially purified protein preparation. If co-fractionation is demonstrated between 3 or more proteins, then add them as a complex.
