Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Epidermal identity is maintained by cell-cell communication via a universally active feedback-loop in Arabidopsis thaliana.

The transcription factors ATML1 and PDF2 are indispensable for epidermal cell fate specification in Arabidopsis embryos. However, the regulation of these genes, and in particular their relationship with cell-cell signalling pathways, although the subject of considerable speculation, remains unclear. Here we demonstrate that the receptor kinase ACR4 positively affects the expression of ATML1 and PDF2 in seedlings. In contrast, ATML1 ... [more]
