Affinity Capture-Western
An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Patched-1 pro-apoptotic activity is downregulated by modification of K1413 by the E3 ubiquitin-protein ligase Itchy homolog.
The Hedgehog (Hh) receptor Patched1 (PTCH1) opposes the activation of Gli transcription factors and induces cell death through a Gli-independent pathway. Here, we report that the C-terminal domain (CTD) of PTCH1 interacts with and is ubiquitylated on K1413 by E3 ubiquitin-protein ligase Itchy homolog (Itch), a Nedd4 family member. Itch induces ubiquitylation of K1413, reduction of PTCH1 at the plasma ... [more]
