MPE1

YKL059C
Essential conserved subunit of CPF cleavage and polyadenylation factor; plays a role in 3' end formation of mRNA via the specific cleavage and polyadenylation of pre-mRNA; contains a ubiquitin-like (UBL) domain, a RNA-binding zinc knuckle motif and a RING finger domain; both the zinc knuckle and RING finger are required for pre-mRNA binding; possible role in ubiquitination of Pap1p; relocalizes to the cytosol in response to hypoxia

Component of the cleavage and polyadenylation factor I (CF I); CF 1, composed of the CF 1A complex (Rna14p, Rna15p, Clp1p, Pcf11p) and Hrp1, is involved in cleavage and polyadenylation of mRNA 3' ends; interacts with the A-rich polyadenylation signal in complex with Rna14p and Hrp1p; mutant displays reduced transcription elongation in the G-less-based run-on (GLRO) assay; required for gene looping and maintenance of genome stability

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Publication

Efficient mRNA polyadenylation requires a ubiquitin-like domain, a zinc knuckle, and a RING finger domain, all contained in the Mpe1 protein.

Lee SD, Moore CL

Almost all eukaryotic mRNAs must be polyadenylated at their 3' ends to function in protein synthesis. This modification occurs via a large nuclear complex that recognizes signal sequences surrounding a poly(A) site on mRNA precursor, cleaves at that site, and adds a poly(A) tail. While the composition of this complex is known, the function of some subunits remains unclear. One ... [more]

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
