SNF1

CAT1, CCR1, GLC2, HAF3, PAS14, AMP-activated serine/threonine-protein kinase catalytic subunit SNF1, L000001944, YDR477W
AMP-activated serine/threonine protein kinase; found in a complex containing Snf4p and members of the Sip1p/Sip2p/Gal83p family; required for transcription of glucose-repressed genes, thermotolerance, sporulation, and peroxisome biogenesis; involved in regulation of the nucleocytoplasmic shuttling of Hxk2p; regulates filamentous growth in response to starvation; SUMOylation by Mms21p inhibits its function and targets Snf1p for destruction via the Slx5-Slx8 Ubiquitin ligase
Saccharomyces cerevisiae (S288c)

UBI4

SCD2, UB14, ubiquitin, L000002414, YLL039C
Ubiquitin; becomes conjugated to proteins, marking them for selective degradation via the ubiquitin-26S proteasome system; essential for the cellular stress response; encoded as a polyubiquitin precursor comprised of 5 head-to-tail repeats; protein abundance increases in response to DNA replication stress

Affinity Capture-MS

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner is identified by mass spectrometric methods.

Purification and characterization of the three Snf1-activating kinases of Saccharomyces cerevisiae.

Elbing K, McCartney RR, Schmidt MC

Members of the Snf1/AMPK family of protein kinases are activated by distinct upstream kinases that phosphorylate a conserved threonine residue in the Snf1/AMPK activation loop. Recently, the identities of the Snf1- and AMPK-activating kinases have been determined. Here we describe the purification and characterization of the three Snf1-activating kinases of Saccharomyces cerevisiae. The identities of proteins associated with the Snf1-activating ... [more]

Additional Notes

  • #LPPI
  • #LPTM
  • Likely post-translational modification
  • Likely protein-protein interaction
  • The ubiquitin could either be a Snf1 conjugate (post-translational modification) or Snf1 could be binding to a protein that is ubiquitinated

Negative Genetic

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.

Reconstituted Complex

An interaction is inferred between proteins in vitro. This can include proteins in recombinant form or proteins isolated directly from cells with recombinant or purified bait. For example, GST pull-down assays where a GST-tagged protein is first isolated and then used to fish interactors from cell lysates are considered reconstituted complexes (e.g. PUBMED: 14657240, Fig. 4A or PUBMED: 14761940, Fig. 5). This can also include gel-shifts, surface plasmon resonance, isothermal titration calorimetry (ITC) and bio-layer interferometry (BLI) experiments. The bait-hit directionality may not be clear for 2 interacting proteins. In these cases the directionality is up to the discretion of the curator.
