HRT1

HRT2, RBX1, ROC1, SCF ubiquitin ligase complex subunit HRT1, L000004443, YOL133W
RING-H2 domain core subunit of multiple ubiquitin ligase complexes; subunit of Skp1-Cullin-F-box (SCF) that tethers the Cdc34p (E2) and Cdc53p (cullin) SCF subunits, and is required for degradation of Gic2p, Far1p, Sic1p and Cln2p; subunit of the Rtt101p-Mms1p-Mms22p ubiquitin ligase that stabilizes replication forks after DNA lesions; subunit of the Cul3p-Elc1p-Ela1p ubiquitin ligase involved in Rpb1p degradation as part of transcription-coupled repair
Saccharomyces cerevisiae (S288c)

CLN2

cyclin CLN2, L000000358, YPL256C
G1 cyclin involved in regulation of the cell cycle; activates Cdc28p kinase to promote the G1 to S phase transition; late G1 specific expression depends on transcription factor complexes, MBF (Swi6p-Mbp1p) and SBF (Swi6p-Swi4p); CLN2 has a paralog, CLN1, that arose from the whole genome duplication
Saccharomyces cerevisiae (S288c)

Dosage Lethality

A genetic interaction is inferred when over expression or increased dosage of one gene causes lethality in a strain that is mutated or deleted for another gene.

Publication

Cdc53/cullin and the essential Hrt1 RING-H2 subunit of SCF define a ubiquitin ligase module that activates the E2 enzyme Cdc34.

SCFCdc4 (Skp1, Cdc53/cullin, F-box protein) defines a family of modular ubiquitin ligases (E3s) that regulate diverse processes including cell cycle, immune response, and development. Mass spectrometric analysis of proteins copurifying with Cdc53 identified the RING-H2 finger protein Hrt1 as a subunit of SCF. Hrt1 shows striking similarity to the Apc11 subunit of anaphase-promoting complex. Conditional inactivation of hrt1(ts) results in ... [more]
