POL1

CDC17, CRT5, HPR3, DNA-directed DNA polymerase alpha catalytic subunit POL1, L000000257, YNL102W
Catalytic subunit of the DNA polymerase I alpha-primase complex; required for the initiation of DNA replication during mitotic DNA synthesis and premeiotic DNA synthesis
Saccharomyces cerevisiae (S288c)

RFX1

CRT1, L000004129, YLR176C
Major transcriptional repressor of DNA-damage-regulated genes; recruits repressors Tup1p and Cyc8p to their promoters; involved in DNA damage and replication checkpoint pathway; similar to a family of mammalian DNA binding RFX1-4 proteins
Saccharomyces cerevisiae (S288c)

Dosage Lethality

A genetic interaction is inferred when over expression or increased dosage of one gene causes lethality in a strain that is mutated or deleted for another gene.

The DNA replication and damage checkpoint pathways induce transcription by inhibition of the Crt1 repressor.

Huang M, Zhou Z, Elledge SJ

We have identified the yeast CRT1 gene as an effector of the DNA damage and replication checkpoint pathway. CRT1 encodes a DNA-binding protein that recruits the general repressors Ssn6 and Tup1 to the promoters of damage-inducible genes. Derepression of the Crt1 regulon suppresses the lethality of mec1 and rad53 null alleles and is essential for cell viability during replicative stress. ... [more]
