Part of actin cytoskeleton-regulatory complex Pan1p-Sla1p-End3p; associates with actin patches on cell cortex; promotes protein-protein interactions essential for endocytosis; binds to and activates Arp2/3 complex in vitro; phosphorylation of Thr-1225 is regulated by MAPK Hog1p in response to osmotic stress; previously thought to be a subunit of poly(A) ribonuclease

Actin assembly factor; C-terminal WCA domain activates Arp2/3 complex-mediated nucleation of branched actin filaments and a polyproline domain which can nucleate actin filaments independent of Arp2/3; mutants are defective in actin cytoskeleton dependent processes such as: endocytosis, bud site selection and cytokinesis; localizes with the Arp2/3 complex to actin cortical patches; homolog of the Wiskott-Aldrich Syndrome protein (WASP), implicated in severe immunodeficiency
Saccharomyces cerevisiae (S288c)

Negative Genetic

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.

We generated a global genetic interaction network for Saccharomyces cerevisiae, constructing more than 23 million double mutants, identifying about 550,000 negative and about 350,000 positive genetic interactions. This comprehensive network maps genetic interactions for essential gene pairs, highlighting essential genes as densely connected hubs. Genetic interaction profiles enabled assembly of a hierarchical model of cell function, including modules corresponding to ... [more]
