SCH9

HRM2, KOM1, serine/threonine protein kinase SCH9, L000001810, YHR205W
AGC family protein kinase; functional ortholog of mammalian S6 kinase; phosphorylated by Tor1p and required for TORC1-mediated regulation of ribosome biogenesis, translation initiation, and entry into G0 phase; involved in transactivation of osmostress-responsive genes; regulates G1 progression, cAPK activity and nitrogen activation of the FGM pathway; integrates nutrient signals and stress signals from sphingolipids to regulate lifespan
Saccharomyces cerevisiae (S288c)
PREY

HOG1

SSK3, mitogen-activated protein kinase HOG1, L000000797, YLR113W
Mitogen-activated protein kinase involved in osmoregulation; controls global reallocation of RNAPII in osmotic shock; activates CDC28 by stimulating antisense RNA transcription; mediates recruitment/activation of RNAPII at Hot1p-dependent promoters; with Mrc1p defines novel S-phase checkpoint that prevent conflicts between DNA replication and transcription; nuclear form represses pseudohyphal growth; autophosphorylates; protein abundance increases under DNA replication stress
Saccharomyces cerevisiae (S288c)

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

The Sch9 kinase is a chromatin-associated transcriptional activator of osmostress-responsive genes.

Pascual-Ahuir A, Proft M

The yeast Sch9 kinase has been implicated in the cellular adjustment to nutrient availability and in the regulation of aging. Here, we define a novel role for Sch9 in the transcriptional activation of osmostress inducible genes. Loss-of-function mutants sch9 are sensitive to hyperosmotic stress and show an impaired transcriptional response upon osmotic shock of several defense genes. We show that ... [more]

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
