ubc-1 encodes an E2 ubiquitin-conjugating enzyme that contains a novel 40 amino acid C-terminal tail, and is homologous to Saccharomyces cerevisiae RAD6/UBC2 which is involved in DNA repair; although loss of UBC-1 activity via RNA-mediated interference (RNAi) does not result in any abnormalities, UBC-1 is able to complement the DNA repair defects of a Saccharomyces cerevisiae rad6 null mutant, suggesting that UBC-1 can function in the DNA repair pathway; ubc-1 mRNA is detectable in embryos.

To verify the genome annotation and to create a resource to functionally characterize the proteome, we attempted to Gateway-clone all predicted protein-encoding open reading frames (ORFs), or the 'ORFeome,' of Caenorhabditis elegans. An interaction is inferred between proteins in vitro. This can include proteins in recombinant form or proteins isolated directly from cells with recombinant or purified bait. For example, GST pull-down assays where a GST-tagged protein is first isolated and then used to fish interactors from cell lysates are considered reconstituted complexes (e.g. PUBMED: 14657240, Fig. 4A or PUBMED: 14761940, Fig. 5). This can also include gel-shifts, surface plasmon resonance, isothermal titration calorimetry (ITC) and bio-layer interferometry (BLI) experiments.
