NPA3

EPA1, GPN1, GTPase NPA3, YJR072C
Member of the conserved GPN-loop GTPase family; has a role in transport of RNA polymerase II to the nucleus; exhibits GTP-dependent binding to PolII; has ATPase activity; involved in sister chromatid cohesion; phosphorylated by the Pcl1p-Pho85p kinase complex; human homolog XAB1 interacts with human RNA polymerase II; protein abundance increases in response to DNA replication stress

Dosage Rescue

A genetic interaction is inferred when over expression or increased dosage of one gene rescues the lethality or growth defect of a strain that is mutated or deleted for another gene.

Npa3 interacts with Gpn3 and assembly factor Rba50 for RNA polymerase II biogenesis.

Liu X, Xie D, Hua Y, Zeng P, Ma L, Zeng F

RNA polymerase II is one of the most vital macromolecular complexes in eukaryotes and the assembly of such complete enzyme requires many factors. Three members of GPN-loop GTPase family Npa3/Gpn1, Gpn2, and Gpn3 participate in the biogenesis of RNA polymerase II with nonredundant roles. We show here that rapid degradation of each GPN protein in yeast leads to cytoplasmic accumulation ... [more]

Reconstituted Complex
Reconstituted Complex

An interaction is inferred between proteins in vitro. This can include proteins in recombinant form or proteins isolated directly from cells with recombinant or purified bait. For example, GST pull-down assays where a GST-tagged protein is first isolated and then used to fish interactors from cell lysates are considered reconstituted complexes (e.g. PUBMED: 14657240, Fig. 4A or PUBMED: 14761940, Fig. 5). This can also include gel-shifts, surface plasmon resonance, isothermal titration calorimetry (ITC) and bio-layer interferometry (BLI) experiments. The bait-hit directionality may not be clear for 2 interacting proteins. In these cases the directionality is up to the discretion of the curator.
