Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Cohesin subunit SMC1 associates with mitotic microtubules at the spindle pole.

Wong RW, Blobel G

Accurate mitotic chromosome segregation depends on the formation of a microtubule-based bipolar spindle apparatus. We report that the cohesin subunit structural maintenance of chromosomes subunit 1 (SMC1) is recruited to microtubule-bound RNA export factor 1 (Rae1) at the mitotic spindle pole. We locate the Rae1-binding site to a 21-residue-long region, SMC1(947-967) and provide several lines of evidence that phosphorylation of ... [more]
