Tuberous sclerosis complex (TSC) is autosomal dominant neurocutaneous disorder caused by TSC1/2 pathogenic variants. We report a child with refractory epilepsy and developmental delay who harbors a de novo TSC2 p.V1646Cfs*7 variant. To elucidate its functional consequences, we assessed the variant's effects on proteostasis, providing a framework for developing therapeutic strategies addressing the underlying molecular disruptions. The child was identified through ... Negative Genetic

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.
