SWI6

PSL8, SDS11, transcriptional regulator SWI6, L000002254, YLR182W
Transcription cofactor; forms complexes with Swi4p and Mbp1p to regulate transcription at the G1/S transition; involved in meiotic gene expression; also binds Stb1p to regulate transcription at START; cell wall stress induces phosphorylation by Mpk1p, which regulates Swi6p localization; required for the unfolded protein response, independently of its known transcriptional coactivators

CLN1

cyclin CLN1, L000000357, YMR199W
G1 cyclin involved in regulation of the cell cycle; activates Cdc28p kinase to promote the G1 to S phase transition; late G1 specific expression depends on transcription factor complexes, MBF (Swi6p-Mbp1p) and SBF (Swi6p-Swi4p); CLN1 has a paralog, CLN2, that arose from the whole genome duplication

Dosage Rescue

A genetic interaction is inferred when over expression or increased dosage of one gene rescues the lethality or growth defect of a strain that is mutated or deleted for another gene.

The E2F functional analogue SBF recruits the Rpd3(L) HDAC, via Whi5 and Stb1, and the FACT chromatin reorganizer, to yeast G1 cyclin promoters.

Takahata S, Yu Y, Stillman DJ

Regulation of the CLN1 and CLN2 G1 cyclin genes controls cell cycle progression. The SBF activator binds to these promoters but is kept inactive by the Whi5 and Stb1 inhibitors. The Cdc28 cyclin-dependent kinase phosphorylates Whi5, ending the inhibition. Our chromatin immunoprecipitation (ChIP) experiments show that SBF, Whi5 and Stb1 recruit both Cdc28 and the Rpd3(L) histone deacetylase to CLN ... [more]
