CKA2

YOR29-12, casein kinase 2 catalytic subunit CKA2, L000000344, YOR061W
Alpha' catalytic subunit of casein kinase 2 (CK2); CK2 is a Ser/Thr protein kinase with roles in cell growth and proliferation; CK2, comprised of CKA1, CKA2, CKB1 and CKB2, has many substrates including transcription factors and all RNA polymerases; protein abundance increases in response to DNA replication stress; regulates Fkh1p-mediated donor preference during mating-type switching

MAF1

RNA polymerase III-inhibiting protein MAF1, L000002614, YDR005C
Highly conserved negative regulator of RNA polymerase III; involved in tRNA processing and stability; inhibits tRNA degradation via rapid tRNA decay (RTD) pathway; binds N-terminal domain of Rpc160p subunit of Pol III to prevent closed-complex formation; localization and activity are regulated by phosphorylation, mediated by TORC1, protein kinase A, and Sch9p; localizes to cytoplasm during vegetative growth and translocates to nucleus and nucleolus under stress conditions

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Casein kinase II-mediated phosphorylation of general repressor Maf1 triggers RNA polymerase III activation.

Graczyk D, Debski J, Muszynska G, Bretner M, Lefebvre O, Boguta M

Maf1 protein is a global negative regulator of RNA polymerase (Pol) III transcription conserved from yeast to man. We report that phosphorylation of Maf1 by casein kinase II (CK2), a highly evolutionarily conserved eukaryotic kinase, is required for efficient Pol III transcription. Both recombinant human and yeast CK2 were able to phosphorylate purified human or yeast Maf1, indicating that Maf1 ... [more]

Biochemical Activity

An interaction is inferred from the biochemical effect of one protein upon another, for example, GTP-GDP exchange activity or phosphorylation of a substrate by a kinase. The bait protein executes the activity on the substrate hit protein. A Modification value is recorded for interactions of this type with the possible values Phosphorylation, Ubiquitination, Sumoylation, Dephosphorylation, Methylation, Prenylation, Acetylation, Deubiquitination, Proteolytic Processing, Glucosylation, Nedd(Rub1)ylation, Deacetylation, No Modification, Demethylation.
