UEV-1

CELE_F39B2.2, mms-2, F39B2.2
uev-1 encodes a ubiquitin-conjugating enzyme (UBC or E2) variant that contains the characteristic UBC motif, but lacks the critical active-site cysteine residue necessary for catalytic activity; UEV-1 activity is required cell autonomously for normal trafficking of the GLR-1 glutamate receptor and consequently, for normal locomotory behavior; UEV-1 physically interacts with the UBC-13 ubiquitin conjugating enzyme that is also required for normal locomotory behavior and that is also likely involved in the DNA damage response; a uev-1::gfp reporter fusion is expressed in a wide variety of tissues including the pharynx, neurons, muscle cells, vulva, intestine, distal tip cell, and cells in the anus/tail; UEV-1 reporter fusions localize to nuclei, the cytosol, and ventral cord neurites.

UBC-13

CELE_Y54G2A.31, Y54G2A.31
ubc-13 encodes an E2 ubiquitin-conjugating enzyme; by homology, UBC-13 is predicted to function in the covalent attachment of ubiquitin molecules to protein substrates, via its association with an E3 ubiquitin-protein ligase; UBC-13 physically interacts with the UEV-1 ubiquitin-conjugating enzyme variant, and like UEV-1, ubc-13 mutations result in defects in GLR-1 glutamate receptor trafficking and hence, abnormal locomotory behavior; in large-scale RNAi screens, loss of ubc-13 activity also results in embryonic lethality and reduced brood size.

To initiate studies on how protein-protein interaction (or "interactome") networks relate to multicellular functions, we have mapped a large fraction of the Caenorhabditis elegans interactome network. Starting with a subset of metazoan-specific proteins, more than 4000 interactions were identified from high-throughput, yeast two-hybrid (HT=Y2H) screens. Independent coaffinity purification assays experimentally validated the overall quality of this Y2H data set. Together ... [more]

Affinity Capture-Western

An interaction is inferred when a bait protein is affinity captured from cell extracts by either polyclonal antibody or epitope tag and the associated interaction partner identified by Western blot with a specific polyclonal antibody or second epitope tag. This category is also used if an interacting protein is visualized directly by dye stain or radioactivity. Note that this differs from any co-purification experiment involving affinity capture in that the co-purification experiment involves at least one extra purification step to get rid of potential contaminating proteins.

Co-fractionation

Interaction inferred from the presence of two or more protein subunits in a partially purified protein preparation. If co-fractionation is demonstrated between 3 or more proteins, then add them as a complex.
