YOS9

YDR057W
ER quality-control lectin; integral subunit of the HRD ligase; participates in efficient ER retention of misfolded proteins by recognizing them and delivering them to Hrd1p; binds to glycans with terminal alpha-1,6 linked mannose on misfolded N-glycosylated proteins and participates in targeting proteins to ERAD; member of the OS-9 protein family
Saccharomyces cerevisiae (S288c)
PREY

IRE1

ERN1, bifunctional endoribonuclease/protein kinase IRE1, L000000875, YHR079C
Serine-threonine kinase and endoribonuclease; transmembrane protein that mediates the unfolded protein response (UPR) by regulating Hac1p synthesis through HAC1 mRNA splicing; role in homeostatic adaptation to ER stress; Kar2p binds inactive Ire1p and releases from it upon ER stress
Saccharomyces cerevisiae (S288c)

Negative Genetic

Mutations/deletions in separate genes, each of which alone causes a minimal phenotype, but when combined in the same cell results in a more severe fitness defect or lethality under a given condition. This term is reserved for high or low throughput studies with scores.

Biological membranes are complex, and the mechanisms underlying their homeostasis are incompletely understood. Here, we present a quantitative genetic interaction map (E-MAP) focused on various aspects of lipid biology, including lipid metabolism, sorting, and trafficking. This E-MAP contains ∼250,000 negative and positive genetic interaction scores and identifies a molecular crosstalk of protein quality control pathways with lipid bilayer homeostasis. Ubx2p, ... Additional Notes

  • An Epistatic MiniArray Profile (E-MAP) analysis was used to quantitatively score genetic interactions based on fitness defects estimated from the colony size of double versus single mutants. Genetic interactions were considered significant if they had an S score > 2.5 for positive interactions (suppression) and S score < -2.5 for negative interactions (synthetic sick/lethality).
