SET1

YTX1, histone methyltransferase SET1, KMT2, L000003286, YHR119W
Histone methyltransferase, subunit of the COMPASS (Set1C) complex; COMPASS methylates histone H3K4; Set1p-dependent H3K4 trimethylation recruits Nrd1p, allowing efficient termination of snoRNAs and cryptic unstable transcripts (CUTs) by Nrd1p-Nab3p-Sen1p pathway; modulates histone acetylation levels in promoter proximal regions to ensure efficient Nrd1p-dependent termination; required in transcriptional silencing near telomeres and at silent mating type loci; has a SET domain
Saccharomyces cerevisiae (S288c)

GLC7

CID1, DIS2, type 1 serine/threonine-protein phosphatase catalytic subunit GLC7, DIS2S1, PP1, L000000706, YER133W
Type 1 serine/threonine protein phosphatase catalytic subunit; cleavage and polyadenylation factor (CPF) component; involved in various processes including glycogen metabolism, sporulation, mitosis; accumulates at mating projections by interaction with Afr1p; interacts with many regulatory subunits; involved in regulation of the nucleocytoplasmic shuttling of Hxk2p; import into nucleus is inhibited during spindle assembly checkpoint arrest
Saccharomyces cerevisiae (S288c)

Synthetic Lethality

A genetic interaction is inferred when mutations or deletions in separate genes, each of which alone causes a minimal phenotype, result in lethality when combined in the same cell under a given condition.

The Set1 methyltransferase opposes Ipl1 aurora kinase functions in chromosome segregation.

A balance in the activities of the Ipl Aurora kinase and the Glc7 phosphatase is essential for normal chromosome segregation in yeast. We report here that this balance is modulated by the Set1 methyltransferase. Deletion of SET1 suppresses chromosome loss in ipl1-2 cells. Conversely, combination of SET1 and GLC7 mutations is lethal. Strikingly, these effects are independent of previously defined ... [more]
